New oral drug shows responses across 6 tumor types

An experimental oral cancer drug has shown signs of effectiveness across six types of solid tumors in patients whose cancers had stopped responding to immunotherapy.

The therapy, GRWD5769, was evaluated in combination with cemiplimab in patients with secondary anti-PD-1 resistance or microsatellite stable colorectal cancer without liver metastases, according to a May 22 news release from drugmaker Greywolf Therapeutics.

Researchers reported response rates of 13% to 36% across six expansion cohorts. Patients had received a median of two prior lines of therapy. Durable clinical benefit rates ranged from 18% to 55%, with progression-free survival durations reaching 33 weeks in non-small cell lung cancer, 16 weeks in hepatocellular carcinoma and 33 weeks in microsatellite stable colorectal cancer without liver metastases.

The study included patients with non-small cell lung cancer, urothelial carcinoma, hepatocellular carcinoma, microsatellite stable colorectal cancer without liver metastases, squamous cell carcinoma of the head and neck and cervical cancer.

The therapy was well tolerated, with no safety signals identified and most adverse events classified as grade 1. The ongoing Emitt-1 trial is being conducted across 28 centers in Australia, France, Spain and the United Kingdom.
