Lilly oral GLP-1 outperforms oral semaglutide in diabetes trial

Eli Lilly’s oral GLP-1 therapy Foundayo (orforglipron) has outperformed oral semaglutide on blood sugar control and weight loss in a Phase 3 trial of adults with Type 2 diabetes.

In the Achieve-3 study, Foundayo 17.2 milligrams delivered a 57.1% greater relative reduction in A1C and a 73.6% greater relative weight loss than oral semaglutide 14 milligrams, according to a June 8 Eli Lilly news release. The trial is the first head-to-head Phase 3 study comparing two oral GLP-1 therapies for Type 2 diabetes.

At 52 weeks, patients taking Foundayo 17.2 milligrams achieved an average A1C reduction of 2.2% compared with 1.4% for oral semaglutide 14 milligrams. Patients receiving Foundayo 17.2 milligrams also lost an average of 19.7 pounds, compared with 11 pounds for oral semaglutide 14 milligrams.

Lilly also reported results from the Achieve-2 and Achieve-5 Phase 3 trials. In Achieve-2, Foundayo lowered A1C by up to 1.7% compared with 0.8% for dapagliflozin at 40 weeks. In Achieve-5, Foundayo lowered A1C by up to 2.1% compared with 0.8% for placebo added to insulin glargine.

The drugmaker said it plans to submit Foundayo for Type 2 diabetes to the FDA by the end of the second quarter.
